摘  要:目的为研究色素上皮相关疾病的发病机制及探讨中医中药治疗此类疾病的作用机制提供细胞模型。方法用胰蛋白酶消化法获得人视网膜色素上皮细胞,免疫细胞化学法对其进行鉴定。采用超低温冷冻的方法对细胞进行冻存,复苏后观察细胞活性。结果人视网膜色素上皮细胞在体外生长良好,胞浆内充满了棕黑色色素颗粒,随着传代的增加,色素颗粒逐渐减少至消失。冻存后复苏的人视网膜色素上皮细胞生长良好,增生活跃。结论成功建立了人视网膜色素上皮细胞的体外培养模式。冻存的细胞复苏后细胞活性不受影响。OBJECTIVE To establish the model of human retinal pigment epithelium(RPE) cells in vitro culture and to study mechanism of RPE disease and effects of traditional Chinese medicine by the model. METHODS Human primary RPE cells were dealt with enzyme digestion and cultivated in vitro culture, then the cells were identified by im- munocytochemical method.The cells were cryopreserved and then observe the cytoactive of frozen cells after thaw- ing. RESULTS RPE cells maintain biological activities in vitro culture. The cells were full of brownish - black pigment granule and when time went on, the brownish - black pigment granule in the cells decreased and even disappeared. Cryopreservation of RPE cells can maintain biological activities of cells with normol growth and features after thawing. CONCLUSIONS The model of human RPE cells cultivated in vitro culture was established successfully. The biological activities of cells were not effective after thawing.
