IL-12基因重组腺病毒对不同肿瘤的抗瘤作用比较

摘　　要：目的:观察比较AdCMVmIL-12的抗肿瘤作用效果。方法:体外用AdCMVmIL-12预处理大鼠甲状腺癌细胞(rMTC),观察其致瘤性;制备动物肿瘤模型,在瘤体内注射AdCMVmIL-12,测量肿瘤体积大小变化。结果:体外用AdCMVmIL-12预处理的rMTC失去致瘤性,不能在动物体内发展成肿瘤;瘤体内注射Ad-CMVmIL-12后,肿瘤体积逐渐缩小。注射后的第12天AdCMVIL-12对rMTC、RTC-R2、H22、XTC-1的抑制率分别为67%、66%、90%、6.4%,与对照组比较差异有统计学意义。结论:AdCMVmIL-12具有明显的抗肿瘤作用,而且对不同类型的肿瘤作用效果不同,具有选择性。Objective： To Observe and compare the anti - tumor effects of AdCMVmlL - 12 vector. Methods： Rat medullary thyroid carcinoma cells（rMTC） were transduced with AdCMVmlL- 12, AdCMVLuc respectively, the transduced cells were washed with DMEM serum - free medium and injected subcutaneously into the flank of the Wistar rats and the tumorgenicity were compared ; Building up four kinds of tumor models with rMTC, RTC - R2, H22,XTC- 1. AdCMVmlL- 12, AdCMVLuc were injected into the tumors respectively and the tumor sizes were compared. Results： The rMTC cells transduced with AdCMVmIL- 12 lost tumorigenicity; The tumor size become much smaller than the control group after it was injected AdCMVmlL - 12. The inhibiting rates of AdCMVmlL - 12 to rMTC, RTC - R2,H22, XTC - 1 were 67% ,66% ,90% ,6.4% respectively. Conclusion： AdCMVmlL - 12 can inhibit the tumor effectively and the inhibiting effect is different in different kinds of tumor.
