构建TGF-βⅡR靶向siRNA真核表达载体并转染人T细胞的研究

摘　　要：目的:构建TGF-βⅡR靶向siRNA真核表达载体并转染T细胞,为进一步探索TGF-β、T细胞与牙周病的关系奠定基础。方法:以TGF-βⅡR为靶基因构建siRNA真核表达载体,转染成功分离培养的人T细胞,转染结果经RT-PCR和Western blot检测。结果:酶切鉴定正确的克隆测序结果与设计的目的序列完全一致,转染siRNA1的T细胞组的TGF-βⅡR mRNA的表达被明显抑制。结论:成功构建了TGF-βⅡR靶向siRNA真核表达载体并成功转染T细胞。AIM： To design and establish siRNA eukaryotic expression vectors of recombinant targeting gene TGF-βⅡR, and to transfect them into T cell. METHODS ： Recombinant was designed and established by targeting gene TGF-βⅡR based on TGF-βⅡR cDNA sequence. Three pairs of oligonucleotides were synthesized and inserted into plasmid pSilencer 3.1 H1 to generate siRNA eukaryotic expression vectors. Then, the plasmids were transfected into human T cells and the results were identified with RT - PCR and Western blot. RESULTS ： The recombinant vectors were established successfully and the expression of TGF-βⅡR of T cell which was transfected with the siRNA1 was inhibited obviously. CONCLUSION ：siRNA eukaryotic expression vectors of recombinant targeting gene TGF-βⅡR has been established and it has been transfected into T cell successfully.
