摘  要:目的探讨整合素β1(integrinβ1)、层粘连蛋白(LN)、基质金属蛋白酶9(MMP-9)在前列腺癌组织中的表达及其与前列腺癌侵袭转移的关系,以及它们之间的相关性分析。方法采用免疫组化SP法分别检测integrinβ1、Laminin、MMP-9在15例良性前列腺增生及39例前列腺癌组织中的表达。结果在良性前列腺增生和前列腺癌组织中integrinβ1蛋白的阳性表达分别为80.00%和46.15%;LN的连续性分布(Ⅰ级)的表达率分别为100.00%和35.90%;MMP-9阳性表达率分别为33.33%和82.05%。三种蛋白在良性前列腺增生和前列腺癌组织中的表达差异均有显著性(P<0.05,P<0.01)。三种蛋白的阳性表达均与肿瘤的临床分期有关(P<0.05,P<0.01),但都与前列腺癌的组织学分级无关(P>0.05)。MMP-9与LN之间的阳性表达呈正相关(r=0.580,P<0.01)。结论integrinβ1、Laminin、MMP-9可能共同参与了前列腺癌的浸润转移过程;integrinβ1的低表达、LN的表达级别增高及MMP-9的高表达可作为早期识别具有高侵袭和转移潜能的前列腺癌及判断其预后的参考指标。[ Objective ] To investigate the expression of integrin β1, Laminin and matrixmetalloproteinase (MMP-9) in human prostatic cancer and their involvement in tumor invasion and metastasis, and to analyse the correlation between the expression of integrinβ1, Laminin and MMP-9. [ Methods] Immunohistochemistry SP method was used to detect the expression of integrin β1 ,Laminin and MMP-9 in 15 cases of benign prostate hyperplasia and 39 cases of human prostatic cancer. [ Results] In 15 cases of benign prostate hyperplasia and 39 cases of human prostatic cancer, the positive expression rates of integrin β1 were 80.00% and 46.15% respectively, and continuous linear positive expression rates of Laminin were 100.00% and 35.90% respectively, and positive expression rates of MMP-9 were 33.33% and 82.05% respectively. All the positive expression rates of integrin 131, Laminin and MMP-9 had significant differences between benign prostate hyper- plasia and prostate cancer, they were correlated with clinical stage (P 〈0.05, P 〈 0.01 ), but all had no correlation with the pathological differentiation (P 〉 0.05 ). A positive relationship existed between MMP-9 expression and LN expression in prostatic cancer (r = 0. 580,P 〈 0.01 ). [ Conclusions] Integrin β1, Laminin and MMP-9 may play important roles in the invasion and mestastasis of prostatic cancer. The decreased expression of integrin β1 protein and over- expression of Laminin and MMP-9 in prostatic cancer are useful makers that can be used to identify high aggressiveness and metastatic potential in prostatic cancer, and can predict the prognosis of prostatic cancer.
