青蒿琥酯对大鼠肾小球系膜细胞增殖的影响

摘　　要：[目的]观察青蒿琥酯(Art)对大鼠肾小球系膜细胞(glomerular mesangial cells,GMCs)增殖的影响并探讨其作用机制。[方法]用不同浓度的青蒿琥酯刺激大鼠肾小球系膜细胞,4-甲基偶氮四唑蓝(tetrazolium salt,MTT)法观察其增殖情况。[结果]与正常对照组相比,LPS具有明显诱导GMC增殖的作用(P﹤0.05或P﹤0.01);青蒿琥酯作用24、48、72 h,GMC增殖水平均可受到抑制(P﹤0.05或P﹤0.01),且随药物浓度增大、作用时间延长,抑制作用增强,高浓度组(60 mg/L)72 h抑制率可达51.18%。[结论]青蒿琥酯能够抑制脂多糖诱导的大鼠肾小球系膜细胞增殖。[Objective]To investigate the effects of artesunate on the proliferation of rats＇ glomerular mesangial cells（GMCs）.[Methods]Mesangial cells were induced with different concentrations of artesunate induced by lipopolysaccharide.Cell proliferation was assessed by MTT spectrometric assay.[Results]The OD value of MTT assay showed that：comparing with control group,LPS could promote the proliferation of GMCs（P﹤0.05 or P﹤0.01）;after induction with Art for 24,48,72 hours,the proliferation level of GMCs was obviously inhibited（P﹤0.05 or P﹤0.01）.The inhibiting effect was increased with higher Art concentration and longer inducing time.The inhibition rate of Art（60mg/L）was 51.18% after 72 hours inducing.[Conclusion]Artesunate can inhibit the division and replication of cells.
