Livin和bcl-2 p53基因在宫颈癌组织中表达相关性的初步研究

摘　　要：目的:探讨新的凋亡抑制基因livin在宫颈癌组织中的表达,及其与bc1-2和p53表达的相关性。方法:应用免疫组织化学链霉菌抗生物素蛋白-过氧化酶连接法(SP法),检测livin、bcl-2、p53基因在lO例正常宫颈组织及57例宫颈癌组织中的表达。结果:Livin基因在正常宫颈组织中不表达,57例宫颈癌组织中,40例表达阳性,占70.1%。Livin基因表达阳性率与宫颈癌的病理分级和临床分期有关。宫颈癌组织中p53蛋白表达阳性、阴性者中,livin基因表达阳性率分别为76.7%(33/43)、21.4%(3/14),两者比较,差异有统计学意义(P<0.05);bcl-2蛋白表达阳性、阴性者中,livin基因表达阳性率分别为87.8%(36/41)、25%(4/16),两者比较,差异有统计学意义(P<0.05)。结论:Livin基因的异常表达而引起的细胞凋亡抑制,在宫颈癌的发生中起一定作用,其表达与宫颈癌组织中p53、bc1-2蛋白的异常表达密切相关。Objective：To study the expression of a novel inhibitor gene of apoptosis,livin in cervical carcinomas and its relationship with the expression of bcl - 2 ,p53 genes. Method： Using streptavidin - biotin perosdase （SP ） method, examined the expression of livin, bcl - 2 and p53 genes in 10 normal cervical tissues and 57 cervical carcinoma tissues. Result： Livin was expressed in 40 of 57 cases cervical carcinomas. In contrast,normal cervical epithelium did not express Livin. Overexpression of livin was related to tumor grade and clinical stage. Livin positive cases were strongly associated with bcl -2 expression（ 33/43,76.7%. versus 3/ 14,21.4%, P 〈 0.05 ） and p53 expression （ 36/41,87.8%. versus 4/16,25%, P 〈 0.05 ）. Conclusion： Apoptosis inhibition by livin,alone or in corperation with bcl -2 and p53 may participate in the onset and progression of cervical carcinoma.
