TGF-β_1、TβRII在子宫内膜样腺癌中的表达及其意义

摘　　要：目的通过检测TGF-β1、TβR II在子宫内膜组织和子宫内膜样腺癌中的表达,分析其与临床病理因素的关系,进一步探讨子宫内膜样腺癌的发生、发展的分子生物学机制及其能否作为预测子宫内膜样腺癌浸润、转移潜能和预后的分子生物学指标。方法采用免疫组化SP法对32例子宫内膜样腺癌、10例正常子宫内膜组织、10例单纯性增生内膜组织、10例复杂性增生内膜组织及10例非典型增生内膜组织共计72例石蜡组织进行TGF-β1、TβR II蛋白表达的检测。结果TGF-β1在子宫内膜样腺癌中的表达显著高于正常子宫内膜组(P(0.01)。TGF-β1表达强度与子宫内膜样腺癌的临床分期(P(0.01)、肌层浸润深度(P(0.01)及淋巴结转移(P(0.05)间的差别具有统计学意义。TβR II在子宫内膜样腺癌中的表达显著低于正常子宫内膜组(P(0.05)。TβR II表达强度与子宫内膜样腺癌的临床分期(P(0.05)、肌层浸润深度(P(0.05)及淋巴结转移(P(0.05)间的差别具有统计学意义。结论TGF-β1在正常子宫内膜向内膜癌转变过程中发挥着重要作用。TGF-β1表达强度与子宫内膜样腺癌的临床分期、肌层浸润深度及淋巴结转移有关。TGF-β1促进子宫内膜样腺癌的发生、发展可能与TβR II突变有关。Objective： To investigate the expression and significance of TGFβ1,TβRII in the endometrioid adenocarcinoma,analyze the relationship between their expression and clinicopathologic,explore the machinery of molecular biology for endometrioid adenocarcinoma,and determine whether they can be recognized as markers to forcast invasion,metastasis and prognosis of endometrioid adenocarcinoma.Methods： By immunohistochemical SP methods in the use of monoclonal antibodies for TGFβ1,TβRII,we examined 32 cases of endometrioid adenocarcinoma,10 cases normal endometrium,10 cases simple hyperplasia,10 cases complex hyperplasia,10 cases atypical hyperplasia and detected the expression of TGFβ1 and TβRII in paraffin-embeded sections of endometrium tissues.Results： The positive rate of TGFβ1 was significantly higher in endometrioid adenocarcinoma than that in normal endometrium（P﹤0.01）.There was a positive relationship of TGFβ1 expression with muscle invasion depth（P﹤0.01）,clinical stage（P﹤0.01） and lymph node metastasis（P﹤0.05）.The positive rate of TβRII was significantly lower in endometrioid adenocarcinoma than that in normal endometrium（P﹤0.01）.There was a negative relationship of TβRII expression with muscles invasion depth（P﹤0.01）,clinical stage（P﹤0.01） and lymph node metastasis（P﹤0.05）.Conclusion： TGFβ1 plays an important role in the process of cancerization of normal endometrium.The expression of TGFβ1 has correlation with clinical stage,muscles invasion depth or lymph node metastasis in endometrioid adenocarcinoma.It promotes endometrioid adenocarcinoma generation and development of TGFβ1 related with the mutation of TβRII.
