摘 要:目的:探讨外源性嘌呤核苷酸对吗啡致神经细胞核苷酸合成代谢降低的影响。方法:大鼠肾上腺嗜铬细胞瘤(PC12细胞)分为对照组、吗啡组、单嘌呤核苷酸(AMP+GMP)组、三嘌呤核苷酸(ATP+GTP)组、吗啡+AMP+GMP组及吗啡+ATP+GTP组。应用RT-PCR法检测各实验组腺苷激酶(AK)和次黄嘌呤-鸟嘌呤磷酸核糖转移酶(HGPRT)基因转录产物含量的变化。结果:吗啡组AK mRNA及HGPRT mRNA表达量(分别为1.330±0.108和1.407±0.141)与对照组(1.874±0.161和1.923±0.155)比较明显降低(P<0.01,P<0.05)。吗啡+AMP+GMP组AK mRNA表达量(1.626±0.171)与吗啡组和对照组比较差异无显著性(P>0.05),HGPRT mRNA表达量(1.796±0.168)高于吗啡组(P<0.05)。吗啡+ATP+GTP组AK mRNA表达量(1.107±0.164)低于对照组(P<0.01),HGPRT mRNA表达量(1.563±0.209)与吗啡组及对照组比较差异无显著性(P>0.05)。结论:外源性补充嘌呤核苷酸能改善吗啡所致的PC12细胞嘌呤核苷酸合成代谢关键酶基因表达降低。Objective To investigate the effect of exogenous purine nucleotides on neuron nucleotide anablism depression. Methods Rat pheoehromoeytoma (PC12 cells) were divided into control, morphine, mono-purine nueleotide (AMP+GMP), tri-purine nueleotide (ATP+GTP), morphine+AMP+GMP and morphine+ ATP+ GTP groups. RT-PCR was used to examine the gene transcripts concentrations of adenosine kinase (AK) and hypoxanthine-guanine phosphoribosyltransferase (HGPRT) in each group. Results The concentrations of AK and HGPRT mRNA (1.330±0.108 and 1.407±0.141 respectively) were decreased in morphine group (P〈0.01, P〈0.05) compared with control group (1.874±0.161 and 1.923±0.155 respectively). The expression level of AK mRNA in the group of morphine+AMP+GMP (1. 626±0. 171) had no difference compared with control and morphine group (P〉0.05), and HGPRT mRNA expression level (1. 796±0.168) was higher than that in morphine group (P〈0.05). The expression level of AK mRNA in the group of morphine+ATP+GTP (1. 107± 0. 164) was lower than that in control group (P〈0.01), and HGPRT mRNA expression level (1. 563 ±0. 209) had no difference compared with control and morphine group (P〉0. 05). Conclusion The exogenous supply of purine nucleotides can ameliorate the down-regulation of morphine on purine nucleotide anabolism key enzyme gene expression in PC12 cells.
