摘  要:采用反相高效液相色谱法,对南方红豆杉组织培养产物中的紫杉醇含量进行定量分析。在C18柱上,以含3%四氢呋喃和7%乙腈的甲醇-水(3:1,V/V),为流动相进行分离,检测波长 254 nm。采用外标法定量,测得紫杉醇在0.04~0.8 mg/L范围内具有良好的线性关系,回收率在98.2%~101.6%之间。该法准确、快速、重现性好,结果令人满意。A new high performance liquid chromatographic method for the determination of taxol in tissue culture of Taxus mairei is described. Taxo1 can be separated on a Sheer C18(5 μm, 5 mm ×150 mm) column at 40°C by using a mobile phase containing 7 % acetonitrile and 3% tetrahydrofuran methanol-water (3: l, V/V) at 0. 6 mL/min flow rate. The samples were quantified with an ultraviolet detector operated at 254 nm. The ca1ibration curve was linear in the range of 0. 04~0. 8 mg/L of taxol with a correlation coefficient of 0. 9996. The relative standard deviation for determination of 0. 4 mg/L taxol was less than l. 8% (n= 6) and the recovery of taxol was 98. 2% ~101. 6 %.
