摘 要:目的:观察胸腺肽α_1对脂多糖刺激后人外周血单核细胞人类白细胞组织相容性抗原-DR(HLA- DR)表达率及淋巴细胞凋亡率的影响。方法:分离、培养正常人外周血单核细胞和淋巴细胞,空白对照组仅加1640培养液,内毒素(LPS)组加LPS刺激,LPS+胸腺肽α_1组先用不同浓度(100、10、1、0.1、0.01μg/ml)胸腺肽α_1预处理细胞,5min后再加入LPS(100 ng/ml)刺激。培养12 h后用流式细胞术检测CD14^+单核细胞HLA-DR表达率和外周血淋巴细胞凋亡率。结果:LPS单独刺激单核细胞时,CD14^+单核细胞HLA-DR表达率[(11.14±5.09)%]较空白对照组[(74.00±3.50)%]明显降低,用胸腺肽α_1预处理后,HLA-DR表达率随着胸腺肽α_1浓度的增高而增高(P均<0.01)。LPS单独刺激淋巴细胞时,淋巴细胞凋亡率[(8.16±0.51)%]较空白对照组[(4.40±0.53)%]明显增加,而用胸腺肽α_1预处理后,淋巴细胞凋亡率随着胸腺肽α_1浓度的增高而降低,胸腺肽α_1浓度为10μg/ml和100μg/ml时,淋巴细胞凋亡率[(4.65±0.75)%和(4.50±0.36)%]与空白对照组比较差异无显著性(P均<0.01)。结论:胸腺肽α_1对LPS引起的人外周血单核细胞和淋巴细胞免疫功能下调有显著抑制作用。Objective: To observe the effect of thymosin α1 on immune function of monocyte and lymphocyte in human peripheral blood stimulated by lipopolysaccharide(LPS). Methods: Monocytes and lymphocytes were isolated from peripheral blood of healthy adults. The cells for test were divided into control (cultivated with medium 1640), LPS stimulation alone (stimulated with 100 ng/ml LPS) and LPS stimulation + thymosin α1 groups. In the last group, different doses (100, 10, 1, 0. 1, 0.01 μg/ml)of thymosin α1 were added followed by LPS(100 ng/ml) challenge. After cultivating for 12 hours, expression of HLA-DR antigen on CD14^+ monocyte and lymphocyte apoptosis rate were determined by direct immunofluorescence with a flow cytometer. Results: HI.A-DR expression was significantly lower in monocytes with LPS stimulation m(11.14± 5.09)%] than that in cells without LPS stimulation [(74. 00±3. 50)%]. After being pretreated with thymosin α1 , HLA-DR expression was significantly increased in a concentration-dependent manner. Apoptosis rate in lymphocytes stimulated by LPS alone [(8. 16±0.51)%] was significantly higher than those in controls [(4.40±0.53)%]. Apoptosis decreased when pretreated with thymosin α1 , which also showed a concentration-dependent manner. NO significant difference in apoptosis rate of lymphocytes was found between 10 μg/ml or 100 μg/ml LPS stimulation and controls. Conclusion: The results suggest that thymosin μ has a significant effect on LPS induced immunosuppression of human monocyte and lymphocyte.
