摘 要:目的:对新鲜和保存人羊膜的超微结构进行观察,从形态学角度探讨人羊膜超微结构与其眼表重建之间的关系。方法:采用扫描电镜、透射电镜和光镜分别对新鲜和低温(-80℃)纯甘油中保存6个月人羊膜的超微结构进行观察。结果:电镜下保存羊膜上皮细胞结构保持完整,分泌颗粒与细胞间连接、微绒毛和微皱褶结构均存在。光镜下保存羊膜仍维持完整立方上皮层,细胞形态整齐,两者基底膜完整。结论:新鲜和保存人羊膜均具有完整的上皮细胞层和基底膜,从形态学角度推测羊膜重建眼表的作用与此有关。Objective :To observe the ultrastructure of fresh and preserved human amniotic membrane and research for the relationship between the ultrastructure of human amniotic membrane and ocular surface reconstruction from morphologic view point. Methods:Transmission electron microscope, scanning electron microscope and optical microscope were used to observe the ultrastructure of fresh and cIyopreserved(-80℃)human amniotic membranes for 6 months in pure glycerin. Results :Preserved human amniotic membrane had the same stucture of regular and intact epithelial cells as the fresh one. Their confor- mation, arrangement, microvilli microplicae, cell junctions and secreted grains still existed even though they had been ciyopreserved for 6 months. Conclusion :The human amniotic membrane cIyopreserved at -80℃ for 6 months in pure glycerin has the same ultrastructure as the fresh one.
