摘 要:目的寻找1种能有效吸附香菇多糖中色素的树脂,并研究pH值、温度、上样量等因素对树脂脱色素的影响,同时初步讨论色素在树脂上的吸附行为。方法采用阳离子交换树脂(JK008)脱除多糖中色素,用苯酚硫酸法测定多糖保留率、考马斯亮蓝法测定蛋白质脱除率,分光光度法测定色素脱除率。采用静态吸附试验和动态吸附试验研究树脂对色素的吸附行为。结果阳离子交换树脂法可使脱色率达到93.4%,脱蛋白率达到86.5%,多糖的保留率达到85%,且条件温和。结论阳离子交换树脂法在脱色的同时还可起到较好的脱蛋白作用,有应用于工业生产的潜力;阳离子树脂对色素的吸附符合Freundlich等温吸附方程。在吸附初期,吸附过程符合Boyd液膜扩散方程。Purpose To select a resin which can adsorb pigment of lentinan efficiently, the influence factors, such as pH value, temperature, elution volume and investigate the adsorption behaviors were studied. Methods A kind of cationion exchange resin (JK008) was firstly used to adsorb lentinan in this work, the lentinan holding rate, deprotein rate and the decoloration rate were determinated by sulfur-phenol acid method, Bradford method and Spectrophotometry method respectively, and the adsorption behaviors were studied by using static and dynamic adsorption methods. Results The decoloration rate, and deprotein rate, and lentinan holding rate were 93.4 % , 86.5 % and 85 % respectively in the method, and it can be used under gentle condition. Conclusion Experimental results show that the method can not only decolor but also deprotein efficiently, so it's potential to apply to industry; and the equilibrium data for the adsorption of pigmentare correlated by Freundlich isotherm equation and in the initial period it obeys the Boyd's film diffusion equation.
