合成SSB/La多肽与DNA共同免疫动物导致DNA抗体产生抑制

摘  要:目的观察SSB抗原与DNA抗原共同免疫动物时不同抗体滴度的变化,研究合成SSB/La多肽与DNA共同免疫动物是否导致DNA抗体产生抑制。方法根据人SSB抗原cDNA编码的氨基酸序列,设计合成214-225位的12个氨基酸多肽,将人工合成多肽与血蓝蛋白(keyhole limpet hemocyan in,KLH)进行耦联,与ds-DNA、SSB联合DNA、KLH、PBS分别作为抗原对5组家兔进行免疫,用ELISA法检测产生的抗体。并取动物心、肝、肾、脾和皮肤进行组织学检查和免疫荧光检测。结果ELISA法检测发现,不同抗原免疫家兔均能诱导相应抗体产生。联合免疫组SSB/La抗体的滴度明显高于单独SSB免疫组,而dsDNA抗体的滴度明显低于单独免疫组。此外,混合免疫组中同一家兔SSB抗体效价显著高于DNA抗体效价。免疫后未发现动物以上器官组织学改变。结论SSB多肽、DNA免疫家兔后抗体产生规律存在差异,合成SSB/La多肽与DNA共同免疫家兔后可导致DNA抗体产生抑制。Objective The aim of the present study is to observe the anti -SSB antibody and anti - dsDNA antibody production profiles following immunization with synthetic SSB peptides alone, DNA alone or co - immunization with these two antigens. Methods SSB 214 -225aa peptide was synthesized by organic chemistry solid - phase peptide syn- thesis. 5 groups of rabbits were immunized with the following antigens : synthetic SSB peptides linked with keyhole limpet hemocyanin (KLH), dsDNA, SSB plus DNA, KLH and PBS. The antibodies produced were determined by ELISA. Histopathology and direct immunofluorescence assay were also performed. Results Anti - SSB and anti - dsDNA antibodies were produced through immunization with SSB peptide and DNA respectively. The titre of anti - SSB antibody in co - immunization group was higher than that in SSB peptide immunization group, while the titre of anti - dsDNA antibody in co - immunization group was lower than that in DNA immunization group. It was also revealed that the titre of anti - SSB antibody was relatively high and that of anti - DNA antibody was relatively low in the Co - immunization groups. PatholOgy showed no morphological or immunological abnormalities in the heart, liver, kidney, spleen and skin tissues. Conclusion Inhibition of anti - DNA - antibody production will be induced by co - immunization with synthesized SSB peptides and DNA, implying that the milder manifestations in some forms of LE maybe, at least partly, associated with the upper - hand formation of anti - SSB antibody.
