摘 要:本文以离体血管收缩功能实验方法,研究了由α_1—AR介导心功能不全及其假手术大鼠主动脉和肾动脉的收缩效应并观察CEC对其作用的影响。结果显示:NE介导心功能不全大鼠主动脉的最大收缩明显低于假手术组大鼠(为假手术组的55%),而肾动脉的最大收缩则与假手术组无明显差异,经CFC处理后,使NE介导心功能不全及假手术大鼠主动脉和肾动脉的pD_2值明显降低,最大收缩效应在两组大鼠主动脉均明显降低,但心功能不全组的降低程度(67±24%)显著低于假手术组( 92±5%)(P<0.05);而对两组大鼠肾动脉的最大收缩无明显影响。提示:(1)心功能不全和假手术大鼠的主动脉主要含α_(1D)—AR,肾动脉主要含义α_(1A)—AR,但心功能不全大鼠主动脉所含的α_(1D)—AR比例明显减少;(2)心功能不全大鼠主动脉α_(1D)—AR亚型易发生下调,而肾动脉α_(1A)—AR亚型不易发生下调。Vasoconstrictive responses induced by α1 - adrenoceptor (α1 - AR) and the effects of chloroethylclonidine (CEC) on α1,-AR mediated vasoconstriction in aortae and renal arteries from cardiac dysfunctional rats(CD) and sham -operation (SO) rats were studied. The results showed that the aortae maximal contraction in CD group induced by α1- AR were significantly lower than that in SO group (55% of SO group), there were no differences in the maximal contraction of renal arteries compared with SO group. Pretreatment with CEC decreased pD2 values and maximal contractions of aortae but the declined - degree in CD were significantly lower than in SO and had no effects on the contraction of renal arteries in CD and SO groups. These results suggested that (1)aortae contained mainly α1D -AR, renal arteries mainly α1A -AR and the percentage of α1D-AR in aortae from CD group were decreased significantly; (2)the α1D-ARs in CD group were desensitized easily while α1A-ARs were not .
