摘 要:目的:探讨大鼠脑外伤后乳酸脱氢酶(LDH)和Caspase-9的表达及其时序性变化,并探讨脑损伤的分子机制。方法:采用改良的自由落体法,制备成脑损伤模型。采用分光光度法检测指标LDH,Caspase-9的活性测定。结果:LDH1h检测到下降,6h降低到最低,从6h后开始升高,到24h达到高峰,之后又开始下降。Caspase-9检测到,1h低表达,3h开始升高,12h达到高峰,1d时开始显著下降。结论:大鼠脑外伤后可诱发LDH,Caspase-9的表达,随着时间的增加呈现一定的变化趋势。Objective. To investigate the rat brain post--traumatic lactate dehydrogenase (LDH) and Caspase--9 expression and timing of their changes, and to explore the molecular mechanism of brain injury. Methods. We used a modified free- fall method to develop the brain injury model, and used spectrophotometric detection indicators, LDH and the caspase- 9 activity assay. Results. LDH was decreased at lh of detection,decreased to the lowest at 6 h, following that it began to rise,and reached a peak at 24 h and then began to decline again. Caspase--9 showed lower expression at lh of detection,began to increase at 3 h,and reached a peak at 12 h and then began to fall. Conclusion.Traumatic brain injury in rats induces LDH and the expression of caspase--9 which shows a change tendency with increasing time.
