摘 要:目的探讨乳腺癌中细胞外信号调节激酶磷酸化表达及其意义。方法采用免疫组织化学和Western blot技术检测乳腺腺病、乳腺纤维腺瘤、原发性乳腺癌病灶中磷酸化细胞外信号调节激酶-2(extracellular signal-regulated kinase 2,ERK2)表达情况。结果免疫组化检测在乳腺腺病、乳腺纤维腺瘤、乳腺癌中磷酸化ERK2蛋白阳性率分别为38.10%(8/21)、66.67%(14/21)、87.50%(84/96),磷酸化ERK2蛋白表达在三种疾病之间有统计学意义(χ^2=25.044,P=0.000);Western blot技术检测乳腺腺病、乳腺纤维腺瘤、乳腺原位癌、乳腺浸润性癌中磷酸化ERK2蛋白表达的平均OD值分别为0.655、0.818、0.882、0.933,乳腺癌中磷酸化ERK2表达量明显高于良性病变,结果与免疫组化检测结果一致;乳腺癌ERK2蛋白表达情况在不同年龄段有统计学意义(χ^2=5.677,P=0.017);乳腺癌ERK2蛋白表达与乳腺癌病理组织学类型、临床分期、生存率无显著相关性。结论在乳腺癌中ERK2磷酸化增强与癌细胞生长活性增加有关,ERK2在判断乳腺良恶性疾病时具有辅助价值。Objective To investigate the expression of phosphorylated extracellular signal- regulated protein kinase 2 (p - ERK2), to reveal the correlation between their expression and clinical - pahtology parameters of breast cancer and to analyze the effect on the pathogenesis and progression of breast cancer. Methods The data of clinic and pathology in 21 cases with adenosis of breast,21 cases with fibroadenoma of breast,and 96 cases with breast caner was respectively collected. All specimens were detected by histopathology, immunohistochemistry technique with antibodies against p - ERK2. The correlation of these factors was analysed. 12 cases of every disease were collected. All every fresh specimens were evaluated by Western blot technique with antibodies against of p - ERK2. Results The positive rate of p - ERK2 expression in adenosis, fibroadenoma and carcinoma of breast was 38.10 % ( 8/21 ), 66.67 % ( 14/21 ) and 87.50 % (84/96), respectively. There was statistical significance between p - ERK2 - positive and p - ERK2 - negative patients in these diseases (χ^2 = 25.044, P = 0.000). The ratio of the average OD value of the p- ERK2 expression detected by Western blot technique in adenosis, fibroadenoma, duct cancer in situ and invasive carcinoma of breast were 0.655,0.818,0.882 and 0.933, respectively. The result of p - ERK2 expression evaluated by Western blot technique was similar as the result which was evaluated by immunohistochemistry technique. There was statistical significance between p- ERK2 - positive and ERK2 - negative patients in different ages (χ^2 = 5.677, P = 0.017). There was not significant difference between ERK2 protein expression with the pathologic type, clinical stages and the survival rate. Conclusion p- ERK2 expression in breast cancer is significantly higher than benign lesions of breast. The factor has an important value on differentiation between the benign and malignant lesions of breast.
