摘 要:目的观察大鼠实验性结肠炎白细胞介素-1β和白细胞介素-10的表达及银杏天宝(EGB)对其影响。方法利用三硝基苯磺酸灌肠制备大鼠实验性结肠炎模型。实验设正常对照组,三硝基苯磺酸模型组,5-氨基水杨酸组(5-ASA,100mg/kg),EGB组(200mg/kg)。4周后,评价结肠组织大体形态和组织学评分,检测MPO和SOD活性,免疫组化和ELISA检测肠组织IL-1β和IL-10表达。结果与正常对照组比较,三硝基苯磺酸模型组大鼠结肠组织大体形态和组织学评分显著增加,MPO活性、肠组织IL-1β蛋白表达水平显著升高;SOD活性、肠组织IL-10产生明显减少。EGB能改善三硝基苯磺酸模型组大鼠结肠组织大体形态和组织学损害,使MPO活性显著下降,同时下调IL-1β蛋白表达水平,升高SOD活性,增加IL-10的产生。结论EGB通过下调肠组织IL-1β表达,上调IL-10的产生发挥抗炎的作用,从而减轻大鼠实验性结肠炎的炎症损伤。Objective To observe the expressions of IL-1β and IL-10 in TNBS- induced colitis in rats and the effects of Ginkgo biloba extract (EGB). Methods The colitis model of rats was established by intracolon enema with 2,4,6 -trinitrobenzene sulfonic acid (TNBS,150 mg/kg). Rats were divided into 4 groups: normal group, model group, 5-ASA group (100 mg/kg) and EGB group(200 mg/kg). The macroscopical and histological changes of the colon were evaluated. The activity of superoxide dismutase(SOD) and myeloperoxidasea (MPO) in the colonic mucosa were determined, respectively. The expressions of interleukin -1β (IL-1β) and IL-10 in the colon tissues of colitis rats were detected by immunohistochemistry and ELISA method. Results The macroscopic and histological damage scores, the activity of MPO and the protein expressions of IL-1β were markedly higher in model group than that in normal group. SOD activity and the IL-10 production of colon tissue in model group were significantly lower than those in normal group. Treatment of EGB improved colonic macroscopical and histological damage score, enhanced colon tissue SOD activities, increased colon IL-10 production, and decreased colon IL-1β expression and colon tissue MPO activities. Conclusion The treatment of EGB was effective for relieving and repairing colonic inflammation and the effects were related to stimulation of IL-10 production and down- regulation of IL-1β expression in TNBS-induced colitis.
