摘 要:目的:制备一种能够替代无水乙醇的,毒性低、弥散性好、可使肿瘤坏死更完全的硇砂提取液,并观察其对小鼠肝癌细胞系H22的作用。方法:60只昆明鼠随机分为3组,每组20只。右肢腋下接种小鼠肝癌H22 1∶4稀释后的腹水0.1ml。成瘤后分别给以生理盐水(NS),硇砂提取液(N),无水乙醇(E)瘤内注射,1次/d,共4次。4天后处死小鼠,剥离肿瘤组织,称重,计算抑瘤率。抑瘤率=(对照组瘤重-实验组瘤重)/对照组瘤重。取标本作病理检测,并作组织化学乳酸脱氢酶(LDH)、琥珀酸脱氢酶(SDH)染色及免疫组化(bcl-2,Bax,c-myc)染色。结果:硇砂提取液有较好的体内抑瘤作用,与无水乙醇比较,作用较优或相当;瘤内注射可使肿瘤体积显著缩小,甚至坏死,组间对照差异有显著性意义。能够抑制与肝癌生长密切相关的乳酸脱氢酶、琥珀酸脱氢酶的表达;免疫组化染色bcl-2,Bax,c-myc与对照组相比差异无显著性意义。结论:硇砂提取液与无水乙醇相比,有着更好的肿瘤抑制效果,更好的弥散性,能够直接杀灭肿瘤细胞,有可能替代无水乙醇用于肝癌等恶性实体瘤的局部治疗。Objective: To prepare a kind of Chinese traditional medicine extract liquid (Sal Ammoniac extract liquid) to substitute for absolute alcohol with low toxicity, good dissemination and can cause tumor necrosis more completely and use it in the experimental treatment to H22 ( mouse liver cancer cell strain) . Methods: Sixty Kunming mice were divided into 3 groups randomly and 20 mice each group. At the right armpit of mice in the three groups were inoculated 0. 1 ml ascites of mouse which had been vaccinated with H22 intraperitoneally by the rate of 1: 4. After the formation of the tumors, conduct the intratumoral injection of normal saline (NS), Sal-Ammoniac extract liquid (N) and absolute alcohol (E) respectively once a day and repeat 4 times totally. 4 days later, kill the mice and dissect the tumors. Weigh the tumors and then calculae the inhibition ratio of the tumor. The inhibition ratio of the tumor = (the weight of control group-the weight of experimental group) /the weight of control group. Get the specimen of tumor and conduct the pathological test, histochemical straining ( lactate dehydrogenase, LDH, succinate dehydrogenase, SDH) and immunohistochemical straining ( bcl-2, Bax, c-myc) . Results: The Sal-Ammoniac extract liquid showed relatively better cancer inhibiting effect in vivo. The Sal-Ammoniac extract liquid showed the same or even better anti-cancer effect compared with absolute alcohol ; In vivo experiment, it notably decreased the volume of the tumor and even led necrosis by intratumoral injection, there was significant difference between experiment group and control group. It can also inhibit the expression of the LDH and SDH which have close relationship with the growth of the liver cancer. There were no significant differences between experiment and control groups in the results of immunohistochemical straining (bcl-2, Bax, c-myc) .
