小剂量顺铂与^(32)P玻璃微球联合应用对裸鼠舌鳞癌移植瘤的生长影响

摘　　要：目的观察腹腔注射小剂量顺铂联合瘤内注射32P玻璃微球(32P-GMS)对裸鼠舌鳞癌移植瘤的抑制影响,探讨小剂量顺铂对32P玻璃微球间质注射内放射的增敏机制。方法将40只荷瘤人舌鳞癌Tca8113细胞裸鼠随机分为4组,每组10只(对照组、单纯顺铂组、单纯32P-GMS组和顺铂联合32P-GMS组),分别处理两周后取出肿瘤称瘤体质量,计算各组抑瘤率。取各组肿瘤组织行透射电镜观察,比较各治疗对肿瘤细胞超微结构的影响。结果单纯顺铂组、单纯32P-GMS及联合治疗组抑瘤率分别为57%、46%和78%。电镜观察联合组细胞死亡率达81%。结论小剂量顺铂联合32P玻璃微球间质注射内放射治疗对裸鼠舌鳞癌移植瘤的生长抑制作用较单独应用有明显增强。[Obsjective] To investigate the inhibitory effect of combination use of low dose of cisplatin and interstitial injection of ^32pGMS on Tca8113 cell implantation tumor growth and the hypersensitive effect of cisplatin on ^32pGMS internal radiotherapy. [Methods] The mice with implantation tumor Tca8113 cell were randomly divided into four groups （contral, Cisplatin therapy,^32pGMS therapy and combination therapy）. Specimens were sectioned two weeks after treatment and weighted. The death rate of tumor cells and the inhibition rate of tumor were calculated respectively. The ultrastructural changes of four groups were observed by transmission electron microscope. [Resuits] The growth of tumor was more slowly in the combination therapy group than that in the simple therapy groups by macrography. The inhibition rate and the death rate of tumor cells of the combination therapy group were significantly higher than those of control group and the other two simple therapy groups （P 〈0.05）. More cell damages were displayed in the combination therapy group than in other groups under the electronic microscope. [Conclusions] A low dose of cisplatin could be used as an effective hypersensitive reagent for the internal radiotherapy. The combination could significantly improve the inhibitory effect on mouse Tca8113 cell implantation tumor.
