摘 要:为探索中度嗜盐菌在高盐有机制革工业废水处理中的应用,解决高盐有机制革废水处理这一难题,从山东省威海市路道口盐场晒盐池盐水中分离一株嗜盐菌株YS-1,对该菌株进行原子力显微镜观察、生理生化测定,全细胞脂肪酸分析、16S rDNA序列同源性分析.研究表明,YS-1菌株16S rDNA序列与Halomonas sp.(AB167061)的亲缘关系最为接近,结合上述其它各项结果确定该菌株为盐单胞菌属(Halomonassp.),属中度嗜盐菌;在SBR反应器中对其进行强化高盐制革废水处理试验,同时结合生物活性碳技术,结果表明,含盐9.3%、CODCr=1738mg/L的高盐制革废水,经168h的CODCr去除率为86%,216h的CODCr去除率为98%,该菌株具有强化高盐制革废水处理的功能,通过分离筛选嗜盐菌强化高盐制革工业废水处理具有可行性,分离筛选的嗜盐微生物与生物活性碳技术相结合构建"嗜盐生物活性碳"技术,在高盐制革废水处理中具有重要作用.图4,参17.A moderately halophilic bacterium YS-1 was isolated from the salty water of a Ludaokou salina of Weihai in China and was subjected to research and studied the application of moderately halophilic bacteria in the hypersaline tanning wastewater treatment. The polyphasic taxonomy study was processed through identifying its morphological features by AFM, researched the features of culture and measured physiologic-biochemical index, analyzed 16S rDNA sequence homology and identified the whole-ceU fatty acids by American MIDI automatic system. Phylogenetic analysis based on 16S rDNA gene sequences showed that the closest relative of the strain is Ha/omonas sp. (AB167061). Therefore,on the basis of polyphasic taxonomic data present, a novel species of the Halomonas sp. is proposed. Combined it with BAC technology and applied them to experiments of intensifying hypersaline tanning wastewater treatment processing in the SBR, a significant result is demonstrated. The wastewater salt content of initial wastewater is 9.3% and initial CODCr was 1 378 mg/L. After 168 h's processed, the removal efficiency of CODCr of tanning had achieved 86%, and after 216 h, it achieved 98%.The experiments also showed that YS-1 has foundation in the hypersaline tanning wastewater treatment and present the feasibility of acclimating of halophilic bacteria and running of processing system. It is important for hypersaline tanning wastewater treatment to combine halophilic bacterium with BAC technology. 4figs., 17refs.
