摘  要:目的观察大鼠前脑缺血再灌注后MMP-9的变化及川芎嗪对其表达的影响。方法用夹闭双侧颈总动脉30min的方法,制备缺血再灌注大鼠模型,应用明胶酶谱法(SDS-PAGE enzymograph)测各组脑组织不同时间点MMP-9的表达。结果假手术组各个时间段大鼠前脑组织少量表达MMP-9;缺血再灌注6h组大鼠脑组织即有MMP-9的表达,缺血再灌注24h组、48h组大鼠脑组织大量表达MMP-9;缺血再灌注3d、5d组大鼠脑组织亦可见MMP-9的表达,但较缺血再灌注24h、48h组相比MMP-9表达减少;除川芎嗪6h组不具统计学意义外,其它时间段川芎嗪治疗组与缺血再灌注组相比MMP-9含量均降低。结论脑缺血再灌注后MMP-9的表达可上调;川芎嗪对大鼠前脑缺血再灌注后对MMP-9的表达有干预作用。Objective To observe the variation of MMP-9 after cerebral ischemic-reperfusion and the influence of tetramethylpyrazine on MMP-9. Methods To restore the blood of bilater carotid arteries of rats after 30mins' occlusion. After the gelatinase spectral method (SDS-PAGE enzymograph),the expression of MMP-9 in brains organization was detected at different time of each group. Results The expression of MMP-9 was low in procerebrum at different phases in sham operated group. In the ischemic-reperfusion group, the expression of MMP-9 at 6h and the expression was conspicuous at 24h and 48h. The expression of MMP-9 lasted 5d after ischemic-reperfusion,but it was lower than at 24h and 48h. The content of MMP-9 degraded in all subsets of therapy group than the corresponding phase of ischemic-reperfusion group except the expression at 6h subset. Conclusion The expression of MMP-9 is up-regulated after cerebral ischemic-reperfusion. Tetrametbylpyrazine may interfere the expression of MMP-9 after ischemic-reperfusion.
