摘 要:目的:建立高效液相离子对色谱法测定蒜氨酸的含量。方法:采用 Lichrospher C_(18)色谱柱(4.6 mm×150mm,5μm),流动相为甲醇-磷酸盐缓冲液(0.28%磷酸二氢钾-0.2%庚烷磺酸钠溶液,用85%磷酸调节 pH 为2.1)(10:90),流速为1 mL·min^(-1),紫外214 nm 波长检测,柱温为30℃。结果:蒜氨酸及其有关物质色谱保留时间适宜,分离良好;蒜氨酸在1.0~250μg·mL^(-1)范围内,线性关系良好(r>0.999),日内、日间精密度良好(RSD<1.5%,n=5)。结论:本方法可准确、快速地测定蒜氨酸含量及有关物质检查。Objective: A specific ion - pairing HPLC method was developed for the determination of alliin. Methods: Analysis was carried out on a Lichrospher C18 column(4.6 mm × 150 mm,5μm) with the mixture of methanol and phosphate buffer solution (0.28% KH2PO4 - 0.20% CHs ( CH2 ) 6 SOs Na, pH adjusted to 2.1 with 85% Hs PO4 ) ( 10 :90) as mobile phase and the flow rate was 1.0 mL .min^-1 and UV detection at 214 nm. Results:The retention and resolution of alliin with its related compounds were good. And linear calibration was found for alliin within the range of 1.0-250 μg . mL^-1. The within- day and between- day precision were good with RSD less than 1.5% (n = 5 ). Conclusion: The method is simple, accurate and suitable both for assay and related substances test of alliin.
