摘 要:目的:探讨温胆汤改良方含药血清对AD细胞模型JNK、c-jun磷酸化水平的影响。方法:经老化处理的Aβ25-35(终浓度为5μmol/L)与NG108-15细胞共同孵育24h建立AD细胞模型,运用中药血清药理学方法,以温胆汤改良方含药血清作用于AD细胞模型,倒置相差显微镜观察细胞形态,MTT法检测细胞活性,流式细胞术检测细胞凋亡,SABC免疫组化法检测p-JNK、p-c-jun蛋白的表达。结果:温胆汤改良方能显著性降低NG108-15细胞JNK、c-jun的表达,降低细胞凋亡率。与模型组比较,抑制剂组和含药血清组JNK、c-jun的表达降低有显著性意义(P<0.05)。结论:温胆汤改良方保护由Aβ25-35引起的NG108-15细胞损伤作用,可能与其作用于JNK信号转导通路有着密切的关系。Objective: To investigate the effect of Modified Wendan Decoction (MWD) on expression of p- JNK and p-c-Jun in cellular model of AD. Methods: NG108-15 cells were exposured to 5μmol/L Aβ5-35 for 24h to establish the cell model. After treating with contained-MWD serum, cells morphous were observed, survival rate was tested by MTT and cell apoptosis by FCM, the expression of p-JNK and p-c-Jun was measured by method of immuocytochemistry. Results: MWD obviously reduced the expression of p-JNK and p-c-Jun and cell' s apoptotic rate. Compared with the model group, the expression of p-JNK and p-c-jun in the SP600125 group and the group treated with contained-MWD serum decreased significantly (P 〈 0.05). Conclusion: The mechanism of M WD protecting the NG108-15 cells from injuring by Aβ25-35 maybe relate to the JNK signal pathway.
