摘  要:目的:探讨人参皂苷Rg3对人类肝癌细胞在裸鼠体内生长和转移的抑制作用及其相关机制。方法:构建裸鼠肝癌高转移模型LCI-D20,共21只荷瘤裸鼠,分为3组,即人参皂苷Rg3组、5-FU对照组和生理盐水对照组,每组7只,于肝移植瘤块后第11天开始用药,人参皂苷Rg3组采用灌胃给药,5-FU对照组和生理盐水对照组采用腹腔注射,每日1次,连续3周,于停药后次日解剖裸鼠,观察人参皂苷Rg3对裸鼠肝移植瘤的抑制作用以及对肝、肺转移的影响;通过免疫组化SP法观察人参皂苷Rg3对裸鼠肝移植瘤MMP-2、nm23、CD44和VEGF基因蛋白表达的影响。结果:人参皂苷Rg3组和5-FU对照组的瘤重抑制率分别为23.66%和62.31%,与生理盐水组比较,前者无显著性差异(P>0.05),后者则差异明显(P<0.01);人参皂苷Rg3组肝、肺转移灶的数量较生理盐水对照组为少,但未达到统计学差异(P>0.05)。免疫组化结果显示,人参皂苷Rg3组的nm23和CD44的表达率分别为100.00%和71.42%,与生理盐水组比较均有统计学意义(P<0.05),MMP-2、VEGF的表达率均为85.71%,与生理盐水组比较无统计学意义(P>0.05)。结论:人参皂苷Rg3对裸鼠肝癌高转移模型LCI-D20具有抑制肝癌细胞生长和转移的作用,但作用较弱,其机制可能与调节nm23和CD44的表达有关,需要作进一步深入的研究。Objective:To observe inhibitory effect of ginsenoside Rg3 injection on human hepatocarcinoma cell's growth and metastasis in nude mice and its relative mechanism. Methods:Established an animal model of the nude mice with the high metastastic human hepatocarcinoma cell, which was named of LCI-D20. Twenty-one LCI-D20 nude mice was divided into 3 groups: Rg3 group (20mg·kg^-1·d^-1 5-FU control group (20mg·kg^-1·d^-1 NS control group,7 mice in each group. Experiment started on the 11^th day after the tumor transplantation. Rg3 group nude mice were treated with a method of stomach-lavaging, and the other group nude mice were treated with abdominal injection per day for 3 weeks, then killed and dissected. Inhibitory effect of Rg3 on the transplantation tumor in nude mice and its metastasis of the liver and lung was observed. The changes of MMP-2, nm23, CD44, VEGF's expression and tumor microvesse density (MVD) of the transplanted HCC tissue were investigated by using immunohistochemical method. Results:The inhibitory rate of Rg3 group and 5-FU control group was 23.66% and 62.31%, respectively. 5-FU control group had significant difference with NS control group (P〈0.01),but Rg3 group had no significant difference with NS control group (P〉0.05). Rg3 group had no significant difference with NS control group in the inhibitory rate to the transplanted tumor's metastasis in the liver and lung, including the number of metastasis focus (P〉0.05). The immunohistochemical result showed that the expression rate of CD44, nm23 in Rg3 group was 71.42%and 100.00%,which had significant difference comparing with the NS group(P〈0.05). The expression rate of MMP-2,VEGF in Rg3 group was 85.71%,which had no significant difference to the NS group(P〉0.05). Conclusion:In the LCI-D20 model, ginsenoside Rg3 injection can inhibit the growth and metastasis of hepatocarcinoma cell, though the effect is not strong.
