摘 要:目的探讨胶质瘤中垂体瘤转化基因(PTTG)、周期素依赖性激酶抑制因子P27在胶质瘤中的作用以及它们与胶质瘤细胞周期的关系。方法40例胶质瘤标本分为Ⅰ级8例、Ⅱ10例、Ⅲ级13例、Ⅳ级9例,RT-PCR检测PTTG及P27mRNA的表达,免疫组织化学检测PTTG、P27及周期素依赖性激酶4(CDK4)的表达。结果PTTGmRNA在Ⅰ~Ⅳ级的表达分别为(0.907±0.065)、(1.109±0.083)、(1.312±0.089)、(1.499±0.215),组间比较差异有统计学意义(P<0.05或0.01);P27mRNA在Ⅰ~Ⅳ级的表达分别为(1.308±0.117)、(1.105±0.200)、(0.826±0.163)、(0.610±0.063),组间比较差异有统计学意义(P<0.05或0.01);PTTG蛋白及CDK4蛋白表达均随肿瘤病理级别的增高而增加,两者在各组间比较差异有统计学意义(均P<0.05).P27蛋白表达随肿瘤病理级别的增高而降低,组间比较差异有统计学意义(P<0.05);PTTG与CDK4蛋白表达之间呈正相关(P<0.01),与P27蛋白表达之间呈负相关(P<0.01),P27蛋白与CDK4蛋白表达之间呈负相关(P<0.01)。结论PTTG、P27在胶质瘤的发生、发展中起重要的作用,两者的异常表达有细胞周期调控有关。Objective To investigate the role of expressions of pituitary tumor transforming gene(PTTG) and p27 in glioma,and explore the relationship between the expressions of them and cell cycle regulation. Methods Specimens of 40 patient with gliomas were divided into grade Ⅰ :8 cases, grade Ⅱ :10 cases, grade Ⅲ :13 cases, grade Ⅳ: 9 case-s, PTTGmRNA and p27mRNA were detected by reverse transcription polymerase chain reaction (RT-PCR) ,and their expressions of PTTG, P27, CDK4 protein were detected by immunostaining assay using streptavidin-peroxidase(SP) method. Results The expressions of PTTGmRNA in grade Ⅰ- Ⅳ were (0. 907 ±0. 065 ), ( 1. 109 ± 0. 083 ), ( 1.312 ± 0. 089 ), ( 1. 499 ±0.215 ) respectively, there was significant difference anaong the groups(P 〈 0.05 or 0.01 ) ; the expressions of P27mRNA in grade Ⅰ-Ⅳ were ( 1. 308 ±0. 117), ( 1. 105± 0.200), (0. 826±0,163), (0. 610± 0. 063) respectively, there was significant difference among the groups( P〈 0.05 or 0.01 );the expressions of PTTG, CDK4 protein increased with the increasing of pathological grade of glioma, there was significant difference among the groups( P 〈 0.05 ). The expressions of P27 protein decreased with the increasing of pathological grade of glioma, there was significant difference among the groups( P 〈 0.05 ). The expressions of PTTG protein had negative correlation with the expression of P27 protein(P 〈 0.01 ) ,and had positive correlation with the expression of CDK4(P 〈 0.01 ) ;the expression of P27 protein had positive correlation with the expression of CDK4(P〈 0.01).Conclusion The abnormal expressions of PTTG and P27 played an important role in development and progression of glioma,and both of them were cell cycle dependents.
