摘  要:为进一步探讨硫氧还蛋白1(thioredoxin1,Trx1)过表达对高糖环境下肾小球系膜细胞基质金属蛋白酶9(Matrix metalloproteinase9,MMP9)表达的影响.采用RT-PCR和明胶酶谱法检测细胞中MMP-9 mRNA和酶活性的变化,用脂质体介导瞬时转染法转染正义Trx1及反义Trx1,观察高糖环境Trx1过表达对MMP9表达的影响.结果表明,HBZY-1高糖组与正常糖组比较,MMP9 mRNA在12h,24h,48h时表达增加(P<0.05),同时酶活性于12h、24h、48h也明显增高(P<0.01).转染正义Trx1组细胞,MMP-9 mRNA水平及MMP9酶活性,在高糖组与正常糖组差异无统计学意义(P>0.05);但转染反义Trx1组和未转染组细胞的MMP9 mRNA水平及酶活性,高糖组均比正常糖组表达增加,差异有统计学意义(P<0.01).提示Trx1过表达可抑制高糖环境诱导的肾小球系膜细胞MMP9高表达.In order to explore the effects of thioredoxinl overexpression(Trx1) on matrix metalloproteinase 9 (MMP9) in glomerular mesangial cells, mRNA expression level and enzyme activity of MMP9 were examined by reverse transcription polymerase chain reaction (RT-PCR) and Gelatin zymography respectively. To observe the effects of Trx1 overexpression on the expression level of MMP9, sense-Trx1 and anti-Trxl were transfected into the glomerular mesangial cells by liposome-mediated transient transfection. Results showed the expression level of MMP9 mRNA was increased at 12 h,24 h,48 h (P 〈 0.05), and the enzyme activity was also increased obviously at 12 h,24 h,48 h (P 〈 0.01). For sense-Trxl cells, there was no statistical significance on the expression level and enzyme activity of MMP9 between high glucose group and normal glucose group(P 〉 0.05). However, for anti-Trx1 cells and no-transfection cells,mRNA expression level and enzyme activity of MMP9 were increased significantly in high glucose group compared with normal glucose group(P 〈 0.01 ). It could be concluded that Trx1 overexpression could inhibit expression increase of MMP9 induced by high glucose in glomerular mesangial cells.
