摘 要:目的比较2种不同的灭菌方法对人牙周膜成纤维细胞原代培养成活率的影响,寻找较为合适的灭菌方法,旨在提高人牙周膜成纤维细胞原代培养的成活率。方法28例牙齿标本分为5倍抗生素溶液灭菌组15例和次氯酸钠溶液灭菌组13例,2组分别采用5倍抗生素溶液与次氯酸钠溶液对取得的人牙周膜组织进行灭菌后,均采用组织块法培养人牙周膜成纤维细胞,并对细胞进行形态学和免疫组织化学染色鉴定,比较2种不同灭菌方法细胞培养的成活率。结果次氯酸钠溶液灭菌组无1例细胞成功传代,5倍抗生素溶液灭茵组细胞培养成活率为46.7%,且符合人牙周膜成纤维细胞的形态学和免疫学特征。结论利用舍5倍抗生素溶液灭菌效果与次氯酸钠溶液无差别,但相对更利于人牙周膜成纤维细胞的成活。Objective To compare the influence of two different sterilizing methods on the primary culture of human periodontal ligament fibroblasts (HPLFS). Methods One group used sodium hypochlorite solution and another used isotonic Na chloride with 5×antibiotics to sterilize human periodontal ligament, then human periodontal ligament fibroblasts were cultured by tissue explant culture technique. Morphological and immunocytochemical analysis were used to characterize the cell lineage. The achievement ratio of the two methods was compared. Results In the sodium hypochlorite solution group, none of the cells had successful passage. In the isotonic Na chloride with 5×antibiotics group, the culture achievement ratio was 46.7%, and the cells' morphological and immunocytochemical characteristics analyzed by tissue explant culture technique were consistent with those of typical HPLFs. Conclusion The sterilizing effect of sodium hypochlorite solution and isotonic Na chloride with 5×antibiotics had no difference. But isotonic Na chloride with 5×antibiotics was more beneficial for cells growth.
