摘 要:目的研究水通道蛋白-2(Aquaporin,AQP-2)在实验性慢性心力衰竭(Chronic heart failure,CHF)早期(冠脉结扎2周)和晚期(冠脉结扎12周)小鼠肾脏组织中的表达及意义。方法以左冠状动脉结扎建立小鼠CHF模型。光镜下观察冠脉结扎心衰小鼠肾组织病理改变;用免疫组织化学法检测心衰小鼠肾组织AQP-2蛋白表达;用逆转录-聚合酶链反应(RT-PCR)法检测心衰小鼠肾脏组织AQP-2mRNA表达。结果冠脉结扎2周后,肾小管可见水肿改变,结扎12周后,肾小管有明显空泡形成;结扎12周后肾组织AQP-2基因和蛋白表达显著高于结扎2周组和假手术组,其中结扎2周组较假手术组也有显著增高。结论CHF小鼠肾组织存在AQP-2基因和蛋白表达增加,且随着心力衰竭加重表达增强。Objective To study the expression and significance of aquaporin-2 (AQP-2) in the kidney of mouse at early and late periods of experimental chronic heart failure(CHF). Methods Experimental CHF mice models were set up with coronary artery ligation. Pathological change was observed under microscope. The expression of AQP-2 mRNA and AQP-2 protein was analysed with reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemical methods respectively. Results There were swelling changes in nephric tubule at early stage and vacuole formation in renal tubule at late stage of CHF. Compared with sham-operated group, renal AQP-2 gene and protein expression was significantly increased in two periods of CHF groups. Among the three groups, the expression of AQP-2 in late CHF groups was the highest. Conclusion The gene and protein expression of renal AQP-2 in mouse increases with the advancement of chronic heart failure.
