低能离子注入诱变美伐他汀高产菌株的选育

摘  要:以提高美伐他汀的产量为目的,利用100 keV氮离子分4个剂量并辅之以制霉菌素抗性平板筛选美伐他汀高产菌株。结果表明:注入剂量为1×1014 N+/cm2时在10μg/mL制霉菌素抗性平板上的菌株的正变率最高,达到36.7%。筛选到了高抗性突变株CP-116,该菌株的发酵单位比出发菌株高35%,且遗传性能稳定。实验表明离子注入技术对选育美伐他汀高产菌株方面行之有效。The original strain was mutated by means of different dose low energy ion implantation , followed by resistance selection with nystatin in order to screen mevastatin high producing strain . Under the implantation dose of 100 keY, 1×10^14 N^+/cm^2 and on the plate containing 10 μ g/mL nystatin, the positive variation rate of the strain had reached the highest values 36.7%, the HPLC results showed that mevastatin yield of CP-116 was improved by 35%,and its productivity was rather stable through successive transfer of cultures. These results showed that the low energy ion implantation was an effective method for screening mutation of high mevastatin producing strain.
