阿加曲班对实验性大脑内出血后脑组织MMP-9表达变化的研究

摘　　要：目的研究大鼠实验性大脑内出血(ICH)后脑组织基质金属蛋白酶-9(matrix metalloproteinases- 9,MMP-9)的表达在阿加曲班(argatroban)注入前后的变化及意义。方法采用立体定向技术将自体不凝血注入大鼠尾状核区制备不同时间段的大脑内出血模型。另设加药组,术后30min及术后每天给予同等量的阿加曲班(3.0 mg/kg)。免疫组化染色法检测注药前后出血脑组织MMP-9的表达。结果未加药组MMP-9表达的阳性细胞数在出血后2d达到最高峰,之后又下降。加药后MMP-9表达的变化趋势消失(P>0.05),同一时相MMP-9阳性表达的细胞数与加药前无显著性差别(P>0.05)。结论进行阿加曲班治疗后MMP-9表达无显著性改变,MMP-9在出血后脑组织可能具有双重作用。Objective To investigate the influence of argatroban（a kind of direct thrombin inhibitor） no Matrix Metalloproteinases-9（MMP-9） expression in brain tissue at different time after experimental cerebral hemorrhage in rats. Methods Stereotactic apparatus and autologous blood was infused into the left caudate nucleus to build up rat experimental Intracerebral hemorrhage（ICH） model. Treatment with argatroban group was the given epactally argatroban 3.0mg/kg 30min and once a day after opteration. Results The expression of MMP-9 peaked at 2d,then decreased. While treated with argatroban,the express tendency of MMP-9 was disappeared,and the expression of MMP-9 did not significantly down-regulate by argatroban. Conclusion The expression of MMP-9 was not significantly changed by argatroban. MMP-9 may had very different roles in tissue after ICH.
