摘 要:以金针菇F19为出发株,通过制备菌丝断片悬液和紫外线照射(3—14min)来筛选突变株。结果发现,12min处理使菌丝体生物量减少了5.14%(p>0.05),但富锌量提高了18.78%(p<0.01);在连续3代的遗传稳定性实验中,菌丝体生物量为1.342—1.419g/100ml,富锌量为49.78—50.45mg/g,与原代差异均不显著(p>0.05)。PAGE技术测定发现,EST是金针菇遗传分析最适标记之一;高锌通过提高EST、POD的合成和活性来促进有机锌的转化和菌丝体的生长。This experiment took Flammulina Velutipes F19 as the original strain screens the mutant F19 of Flammulina velutipes by preparing mycelium fragment in suspending water and irradiated with ultraviolet rays (3-14min) .It was found that when the irradiation time reached 12min,although the mycelium biomass reduced by 5.14% (p 〉 0.05), the zinc dosage rose by 18.7% (p 〈 0.01). The hereditary stability experiment of successive three generations suggested that the mycelial biomass was 1.342-1.419g/100ml, the zinc dosage was 49.78--50.45mg/g and there was no significant difference between the average biomass and the zinc dosage (p 〉 0.05). Further analysis of mutating Flammulina velutipes with isodynanfic enzyme and the relevant index before and after the zinc enrichment showed that EST was one of the most suitable marks in the hereditary analysis of Flammulina velutipes, the lfigh zinc promoted the conversion of organic zinc and the growth of mycelium by increasing the synthesis and activity of enzyme EST and POD.
