摘 要:目的:观察仙人掌提取物对糖尿病小鼠血糖和氧化应激的影响,以探讨其对糖尿病的治疗机制。方法:小鼠一次性腹腔注射链脲佐菌素(streptozotocin,STZ)溶液150mg/kg建立小鼠糖尿病模型。将小鼠分成4组:正常对照组、模型对照组、仙人掌小剂量组、仙人掌大剂量组。灌胃给予仙人掌提取物1.0g/kg和2.0g/kg,连续4周。实验前、第2周、第4周测空腹血糖;末次给药后2h,断头取血和肝脏组织,分别检测血清和肝脏超氧化物歧化酶(SOD)、丙二醛(MDA),同时测定血清总抗氧化能力(T-AOC)。结果:①空腹血糖:给予仙人掌提取物2周后,大剂量组小鼠血糖明显低于模型对照组(P<0.05),4周后,大、小剂量治疗组均明显低于模型对照组(P<0.05,P<0.01)。②SOD、MDA和T-AOC变化:与高血糖模型对照组相比,大、小剂量治疗组血清和肝脏中SOD活力明显上升(P<0.01),MDA含量均显著降低(P<0.05,P<0.01),血清T-AOC增强(P<0.01)。结论:仙人掌提取物能明显降低糖尿病小鼠血糖,且能降低糖尿病小鼠氧化应激水平。Objective:To observe the effect of the extract of opuntia on the level of blood glucose and oxidative stress in Diabetic Mouse,and study the mechanism that opuntia cures diabetes.Methods:Animal model of diabetes was established by injecting streptozotocin(150 mg/kg) into mice abdominal cavity.The mice were divided into four groups: normal control group, diabetic model group, low-dosage extract group, high-dosage extract group.Blood glucose level was determined with blood glucose monitor before being given the extract of opuntia (1.0 g/kg and 2.0 g/kg ),2 weeks and 4 weeks after being given the extract of opuntia.The mice were killed on the end of 4th week.Activity of SOD and concentration of MDA in blood serum and liver were determined, and T-AOC in the serum was determined.Results:①Fasting blood glucose:After two-week treatment of the extract of opuntia, the fasting blood glucose level in the high-dosage extract group was markedly lower than that in the hyperglycemia model group (P< 0.05),4 weeks later, the blood glucose level in the low-dosage and high-dosage extract groups was markedly lower than the hyperglycemia model group (P<0.05,P<0.01)②SOD activity,MDA content and T-AOC:Compared with the hyperglycemia model group, Both in serum and liver, the SOD activity of the low-dosage, high-dosage extract groups was markedly higher than that in the hyperglycemia model group(P<0.05,P<0.01);MDA content in the serum and liver of the low-dosage extract group was significantly decreased(P< 0.05),and that in the high-dosage extract group was very significantly decreased (P< 0.01);T-AOC in the serum of the low-dosage, high-dosage extract groups was markedly higher than that in the hyperglycemia model group(P<0.01).Conclusions:The extract of opuntia could remarkably decrease the blood glucose level of diabetic mice, the mechanism of the extract of opuntia in reducing the blood sugar may be related to its effects on enhancing the activity of antioxidase and protecting the tis sues from peroxidation damage.
