摘 要:目的:探讨胶质瘤中垂体瘤转化基因(PTTG)和血管内皮生长因子(VEGF)在胶质瘤中的表达以及二者之间的相关性。方法:将40例胶质瘤患者手术标本按病理级别分为级8例,级10例,级13例,级9例,逆转录聚合酶链式反应(RT-PCR)检测PTTG及VEGFmRNA的表达,免疫组化检测PTTG、VEGF蛋白的表达。结果:PTTG蛋白表达在~级的阳性率分别为37.5%,50.0%,76.9%,100.0%,其表达随着肿瘤级别的增高而增加(χ2=9.602,P<0.05);VEGF蛋白~级的阳性率分别为25.0%,40.0%,76.9%,88.9%,二者的表达均随着肿瘤级别的增高而增加(P<0.05)。PTTGmRNA的表达在~级分别为0.907±0.065、1.109±0.083、1.312±0.089、1.499±0.215,组间有极显著性差异(P<0.01);VEGFmRNA的表达在~级分别为1.024±0.118、1.139±0.057、1.415±0.094、1.693±0.128,组间有显著性差异(P<0.01)。PTTG、VEGF蛋白表达呈显著正相关(P<0.01)。结论:PTTG、VEGF的异常表达在胶质瘤中起协同作用,二者共同促进胶质瘤的发生发展。Objective: To investigate the expressions and their relationship of pituitary tumor transforminggene (PTTG) protein and vascular endothelial growth factor (VEGF) in glioma . Methods: Reverse transcription polymerase chain reaction (RT-PCR) was used to detected the expressions of PTTGmRNA and VEGF mRNA in specimens of 40 patient with gliomas which included grade Ⅰ 8cases ,grade 10 cases ,grade Ⅱ 13 cases,grade Ⅳ9 cases. The protein expressions of PTTG and VEGF were detected by immunostaining assay using Streptavidin-Peroxidase (SP) mothed. Results: The positive rate of PTTG protein in grade Ⅰ -Ⅳ was 37. 5% ,50.0%,76.9% ,100.0%respectively, the positive rate of VEGF protein in grade Ⅰ- Ⅳ was 25.0%, 40. 0%, 76.9 %, 88.9 % respectively ,and the protein expression of PTTG and VEGF increased with the increasing of the pathological grade(P〈0.05). The expressions of PTTGmRNA in grade Ⅰ -Ⅳ were 0.907±0.065 ,1. 109±0. 083 ,1.312±0.089, 1.499±0.215 respectively,there was a significant difference between any two groups(P〈0.05 or P〈0.01); the expressions of VEGF mRNA in grade Ⅰ -Ⅳ were 1. 024± 0. 118,1.139 ±0. 057,1. 415±0. 094,1. 693±0. 128 respectively, there was a significant difference between any two groups (P〈0. 05 or P〈 0. 01). The expressions of PTTG protein had positive correlation with the expression of VEGF protein (P〈0.01). Conclusion.. The abnormal expressions of PTTG mRNA and VEGF mRNA and their protein play an important role in development and progression of glioma,and they have positive correlation in glioma.
