摘  要:目的:探讨小鼠肝癌细胞H22培养上清液对鼠L929细胞细胞周期、CyclinD1、P27蛋白表达的影响。方法:采用肝癌细胞H22的培养上清液诱导正常成纤维细胞L929,诱导细胞为L929-H22。采用流式细胞术检测L929细胞与L929-H22细胞细胞周期分布,免疫细胞化学方法检测2种细胞中CyclinD1、P27蛋白的表达。结果:与L929相比,L929-H22G0/G1期细胞比例明显减少,S期细胞比例明显增加(P<0.05),增殖指数PI明显升高(P<0.05);CyclinD1的表达增强(P<0.05),P27的表达减弱(P<0.05)。结论:小鼠肝癌细胞培养上清液诱导的细胞CyclinDl、P27的表达及细胞周期分布与正常细胞不同,CyclinDl表达增强和P27表达减弱可能在细胞恶性转化过程中发挥作用。Aim: To investigate the expression of CyelinD1 and P27 and cell cycle distribution of normal cells and cells induced by the supernatant of mice hepatocellular carcinoma cells and the probable mechanism. Methods :The normal L929 cells were induced by the supernatant of mice hepatocellular carcinoma cells H22 and named the induced cells as L929- H22 cells. The FCM assay was used to detect cell cycle distribution of L929 and L929-H22 cells. The immunocytochemistry assay was used to detect the expression of CyelinD1 and P27 in L929 and L929-H22 cells. Results:Compared with L929 cells, the proportion of L929-H22 cells in S-phase increased while in G0/Gl-phase decreased significantly and the proliferating index was hightened (P 〈 0.05). Compared with L929 cells, L929-H22 cells showed significantly higher expression level of CyelinD1 (P 〈0.05) and lower expression level of P27 (P 〈0.05). Conclusion: The expression of CyclinD1 and P27 and cell cycle distribution of cells induced by supernatant of mice hepatocellular carcinoma cells was different from those of norreal cells. The higher expression of CyclinD1 and lower expession of P27 may play an important role in the carcinogenesis.
