摘 要:目的检测胃癌和癌旁组织RASSF1A基因启动子甲基化改变规律。方法取40例胃癌患者肿瘤组织及癌旁组织,用甲基化特异性聚合酶链反应(MSP)检测RASSF1A基因启动子的甲基化。结果癌旁组织仅观察1例甲基化(2.5%),而肿瘤组织中26例出现甲基化表达(65.0%)。(P(0.05)。结论RASSF1A基因启动子区甲基化是是胃癌经常发生的分子事件,可能与胃癌癌变有一定关系。RASSF1A基因甲基化是对胃癌高危人群进行筛选的一项重要的候选指标。Objective To determine RASSF1A methylation in the cancer and adjacent cancer tissues. Methods Sample primary tumors were collected from 40cases of primary gastric cancer. Adjacent cancer tissues and carcinoma tissues were selected from every subjects. Methylation of RASSF1A gene promoter was detected using methylation - specific PCR (MSP). Results Promoter methylation of RASSF1A could be detected in 26cases(65.0% ) in gastric carcinoma, while only one found in adjacent cancer tissues (2.5 % ) ( P 〈 0. 05 ). Conclusion RASSF1A methylation was the possible mechanism of carcinogenesis of gastric carcinoma and might be a frequent molecular event in gastric carcinoma. Thus, RASSF1A methylation could serve as a useful biomarker to screen the high - risk subjects and to make early diagnosis for gastric carcinoma.
