摘 要:目的研究壳寡糖Ⅰ(聚合度2~10)和壳寡糖Ⅱ(聚合度2~30)对四氯化碳(CCl4)诱导的化学性肝损伤小鼠的保护作用。方法小鼠连续7d按167mg·kg^-1·d^-1灌胃给予两种壳寡糖,并于第7天腹腔注射CCl4,检测血清丙氨酸氨基转移酶(Au)和天门冬氨酸氨基转移酶(AST)活性,肝组织中丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性;采用光学显微镜观察肝脏组织形态学变化?结果两种壳寡糖都能明显抑制小鼠血清ALT和AST活性的升高(P〈0.01),抑制肝组织中MDA含量升高(P〈0.01),提高肝组织中SOD活力(P〈0.01),减轻CCl4对肝脏细胞的病理损伤。结论两种壳寡糖对四氯化碳造成的小鼠急性肝损伤具有一定的保护作用,并且壳寡糖Ⅱ的肝脏保护作用优于壳寡糖Ⅰ。Objective To investigate the protective effect of oligochitosan Ⅰ (degree of polymerization 2 - 10) and oligochitosan Ⅱ ( degree of polymerization 2 - 30) on chemical hepatic injury induced by CCI4 in mice. Methods Two types of oligochitosan were administrated at 167 mg·kg^-1·d^-1 for 7 days and the liver injury model was established by administration with carbon tetrachloride ( CCl4 ). The activities of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST), the contents of malondialdehyde (MDA) and the activities of superoxlde dismutase (SOD) in hepatic tissues were measured. The hepatic histological changes were observed by optical microscope. Results Two types of oligoehitosans remarkably inhibited ALT and AST in serum( P 〈 0.01 ) , and decreased the content of MDA( P 〈 0.01 ), meanwhile increased the activities of SOD in hepatic tissues. Furthermore, the pathological injury was also significantly alleviated in the oligoehitosan groups. Conclusion Two types of oligoehitosans showed protective effect on the acute hepatic injury induced by CCl4, and in which oligoehitosan 1/ was better than oligochitosan Ⅰ.
