摘  要:目的探讨放射后肝癌细胞侵袭潜能的变化及其机制。方法以不同剂量(0、4、8Gy)高能 X 线照射人肝癌低转移潜能细胞株 MHCC97L。明胶酶谱法检测放射后24、48、72、96 h 细胞培养上清液金属蛋白酶(MMP)-2活性,细胞免疫荧光法检测放射后24、48和72 h 细胞内 MMP-2蛋白表达,Transwell 法测定放射后48和96 h 细胞的侵袭能力。结果放射后细胞 MMP-2蛋白分泌及活性增强并与放射剂量及时间相关,8 Gy 放射后48 h MMP-2蛋白活性最高,达到对照组的2倍。0、4、8 Gy 放射后48 h 穿过 transwell 小室的细胞数分别为7.2±1.9、13.2±2.7和31.2±7.2(P<0.05),放射后96 h 组间差异无统计学意义。结论放射后肝癌细胞侵袭潜能短暂增强,放射诱导细胞 MMP-2分泌及活性增强是侵袭性增强的原因之一。Objective To investigate the effects of radiation on the invasion of hepatocellular carcinoma (HCC) and the mechanism. Methods HCC cell line MHCC97L was irradiated at different doses (0, 4, 8 Gy). Matrix metalloproteinase-2 (MMP-2) plotein activity was examined by gelatin zymography at 24, 48, 72 and 96 h, and MMP-2 protein expression was detected by immunofluorescence at 24, 48 and 72 h, Invasion was assessed on matrigel-coated transwell inserts at 48 and 96 h. Results Radiation increased MMP-2 protein secretion and activity of MHCC97L cell line in a time- and dose-dependent man- ner. After radiation with 8 Gy for 48 h, the activity of MMP-2 protein reached the peak and was 2 fold as high as control group. Forty-eight h after radiation at different doses (0, 4, 8 Gy), the number of cells that passed through the transwell inserts was 7.2 ± 1.9, 13.2 ± 2.7 and 31.2 ± 7.2 respectively ( P 〈 0.05), but there was no significant difference 96 h after radiation. Conclusion Radiation transiently increased MMP-2 expression and protein activity of HCC cells and resulted in the enhanced invasive potential of HCC cells.
