摘 要:目的探讨 Hedgehog 信号通路中 SHH 和 PTCH 蛋白在膀胱移行细胞癌中的异常表达及其临床意义。方法采用免疫组织化学方法检测30例膀胱癌及10例正常膀胱黏膜中 SHH和 PTCH 蛋白的表达情况,并分析其阳性率与肿瘤分级分期的关系。结果 SHH 和 PTCH 蛋白表达于膀胱移行细胞癌细胞的胞质,阳性表达率分别为53.33%和60.00%,正常膀胱黏膜中未见两者的阳性表达;SHH、PTCH 蛋白在表浅性膀胱癌的阳性表达率分别为70%和75%,高于浸润性膀胱癌(P<0.05);SHH、PTCH 蛋白在高中分化膀胱癌的阳性表达率分别为68%和79%,高于浸润性膀胱癌(P<0.05)。结论 SHH 和 PTCH 蛋白在膀胱癌中过度表达,两者的阳性率与膀胱癌的分级分期有关,提示 Hh 通路的激活可能部分参与膀胱癌的发生过程。Objective To investigate the abnormal expression and clinical significance of SHH and PTCH protein in human bladder transitional cell carcinoma (BTCC). Methods Immunohistochemistry was used to detect the expression of SHH and PTCH protein in specimen from 30 cases with BTCC and 10 normal bladder mucosa tissues ,the relationships among the positive rate and the tumor grade and stage were analyzed. Results Stain of SHH and PTCH protein was located in the cytoplasm of cancer cell. The positive rates of SHH and PTCH protein in BTCC were 53.33% and 60.00% , respectively. Neither of SHH and PTCH protein expression was detected in normal bladder mucosa tissues. The positive rate of SHH and PTCH protein expression in superficial BTCC was 70%, 75%, respectively, significantly higher than invasive BTCC (P 〈 0.05 ) ;The positive rate of SHH and PTCH protein expression in well-differentiated BTCC was 68 %, 79%, respectively, significantly higher than poorly differentiated BTCC ( P 〈 0.05 ). Conclusion The over expression of SHH and PTCH protein existed in BTCC, the level of protein expression were related with the tumor grade and stage. The activation of Hh signal pathway may participate in the development of BTCC.
