摘  要:目的建立山羊股骨远端骨洞模型,观察万古霉素缓释微球在模型体内释放药物的过程。方法山羊双侧股骨远端形成两个圆柱形骨洞,PMMA 封闭14 d 后,再次暴露骨洞,分别在右侧和左侧移植万古霉素微球和无万古霉素的微球。通过高效液相检测各部位的万古霉素浓度,并以葡萄球菌作为检测菌株,评估其抗菌活性。结果 X 线见骨洞约为2.5 cm×1.5 cm×1.5 cm,移植微球后右侧骨洞内的万古霉素浓度最高,1 d 达最高值(294.71±20.45)mg/L;左侧和血液中的浓度在2 d 达高峰,为(5.54±0.97)mg/L 和(29.98±2.61)mg/L;超过对葡萄球菌敏感折点值(5 mg/L)的持续释放天数分别为21、1、6 d。结论建立的山羊股骨远端骨洞模型适合于观察药物缓释载体在骨组织内释放药物的动态过程,便于检测洗脱出的药物浓度。Objective To establish distal femur bone cavity animal model in goat and observe drug release course from vaneomycin alginate beads coated with fibrin gel ( Vaneo-AB-FG ) transplanted in bone cavity. Methods Two cylindric bone cavities were formed in goat distal femur at both sides. After 14 days' seal by PMMA,the right and left bone cavities were exposed again and filled with alginate beads with vaneomyein and without vaneomyein respectively. The vaneomyein concentration in different sites was assayed by high performance liquid chromatography (HPLC), and its activity test to staphylococcus aureus ATCC25923 was determined using tube dilution method. Results The real size of bone cavity was approximately 2.5 cm× 1.5 cm ×1.5 cm through radiography. The vaneomyein concentration in right cavity reached the peak value on the 1st day after implantation (294.71 ±20.45) mg/L,and the highest values of the left cavity and blood appeared on the 2nd day, (5.54 ±0.97 ) mg/L and (29.98±2.61 ) mg/L respeetively. The vaneomyein concentration in right cavity,left cavity and blood was above the breakpoint sensitivity concentration for 21,1 and 6 days respectively. Conclusion The distal femur bone cavity animal model in goat established by authors can be used to observe the release course of drug in bone tissue, especially detect drug concentration eluted from drug carriers.
