摘  要:目的观察颞下颌关节盘前移位动物模型的滑膜组织中血管内皮细胞生长因子(VEGF)表达水平的变化,探讨其在关节盘前移位致病机制中的作用。方法10只4~6月龄日本大耳白兔,随机分为正常对照组(2只),手术和手术对照组(8只),手术使颞下颌关节盘前移位;术后1、2、4、8周分别处死2只动物;免疫组织化学方法检测滑膜组织中 VEGF 的表达。结果正常对照组以及对照侧滑膜组织 VEGF 表达呈阴性,实验侧滑膜组织术后 VEGF 呈阳性,并且 VEGF 的表达随时间发生改变。结论颞下颌关节盘前移位滑膜组织中 VEGF 表达增强,说明其可能参与了这种病症的发病机制。Objective To investigate the change of vascular endothelial growth factor (VEGF) expression and its significance in the synovium after anterior disc displacement. Methods Ten healthy adult Japanese rabbits were used in this study. The temporomandibular joint disc was surgically displaced anteriorly in one side joint in each of 8 rabbits while the other side treated with sham-operation and two rabbits without operation were as control. Two rabbits were sacrificed 1,2,4 and 8 weeks post-operation respectively. The expression of VEGF in the synovial membrane of the animal models was examined by SP immunohistochemical staining. Results Experimental and sham-operative animals tolerated the surgical procedures well. The discs displaced anteriorly were observed in the by general observation. The expression of VEGF was detected mostly in the plasma of synoviocytes, vessel endothelial cells and fibroblasts of the experimental group while it was not presented in the control groups. Conclusion The expression of the VEGF in the synovium of the experimental group indicated that the VEGF might play an important role in the development and prognosis of the temporomandibular alteration after anterior disc displacement.
