类胰蛋白酶对肝星状细胞增殖及Ⅰ型胶原mRNA表达的影响

摘　　要：目的观察类胰蛋白酶对肝星状细胞(HSC)增殖及Ⅰ型胶原mRNA表达的影响,探讨类胰蛋白酶在肝纤维化中的作用。方法用Percoll密度梯度离心法分离、培养大鼠HSC,经系列浓度类胰蛋白酶作用后,分组进行如下实验:用噻唑兰比色法(MTT)检测HSC的增殖;用RT-PCR法检测Ⅰ型胶原mRNA的表达。结果肥大细胞(MC)类胰蛋白酶(1～100ng/ml)能够促进HSC增殖及Ⅰ型胶原mRNA的表达,此作用呈剂量依赖性(各实验组与对照组比较,P<0.05)。结论肥大细胞类胰蛋白酶可直接作用于HSC,参与肝纤维化过程。Objective To observe the effects of mast cell tryptase on the proliferation of HSC and the expression of collagen Ⅰ in hepatic stellate cell （HSC） and to study the role played by tryptase during development of liver fibrosis. Methods HSC were isolated from rat liver with percoll and treated with an increasing amount of tryptase and divided into groups for further experiments. Proliferation of HSC was measured by means of Methyhhio tetrazole （MTI） method. The expression of collagen Ⅰ mRNAs in vitro cultured HSC was assessed by semi-quantitive reverse transcriptase polymerase chain reaction （RTPCR）. Results Mast cell tryptase （1-100ng/ml） promoted proliferation of HSC and expression of collagen Ⅰ mRNA of HSC in a dose-dependent manner （P〈0.05）. Conclusion Tryptase is involved in occurrence and development of liver fibrosis by affecting HSC directly.
