摘 要:目的:探讨大肠癌PARP与微血管形成的关系及其可能机制.方法:应用SP免疫组化染色观察30例大肠癌中PAR的表达和微血管密度的变化,流式细胞计数和明胶酶谱观察5-AIQ抑制CT26细胞后MMP-9阳性细胞数和活性的变化.结果:在PAR强表达的大肠癌中,微血管密度高,弱表达则密度低,其差异具有统计学意义(P=0.0001,P<0.01);流式细胞计数中,与未处理组相比,用500μmol/L5-AIQ处理后,MMP-9阳性细胞的百分比降低;明胶酶谱显示,MMP-9的活性在500μmol/L5-AIQ处理组和未处理组中差异有统计学意义(P=0.0013,P<0.01).结论:大肠癌PARP的表达可能和微血管形成相关,PARP活性增强可能促进了肿瘤血管的形成.AIM: To study the relationship between poly (ADP- fibose) polymerase (PARP) and microangiogenesis in colorectal carcinoma and its underlying mechanism. METHODS: Immunohistochemical SP staining was used to examine the expression of poly(ADP-ribose) (PAR) and the change of microvessel density (MVD) under light microscope in 30 cases of colorectal carcinoma. The positive rate and activation of MMP-9 were detected by Flow Cytometry (FCM) and gel zymography after the inhibition of CT26 cells by 5-aminoisoquinolinone ( 5-AIQ ). RESULTSMVD of the group with positive PAR was higher than that of nega- tive PAR, and the difference was significant ( P = 0. 0001, P 〈 0.01 ). The positive rate of MMP-9 decreased after the treatment of 500 μmol/L 5-AIQ. The activation of MMP-9 in 5-AIQ-treated CT26 was significantly weaker than that in 5-AIQ-untreated cells ( P = 0. 0013, P 〈 0.01 ). CONCLUSION : The expression of PARP is correlated with microangiogenesis, and it probably promotes the angiogenesis in colorectal carcinoma.
