家蚕微卫星标记的荧光原位杂交研究

摘  要:家蚕既是重要的农业经济昆虫,又是遗传学研究常用的实验动物和现代分子生物学研究的良好材料.目前已有多种分子标记被用于家蚕遗传连锁图的构建,但人们对这些分子标记在染色体上的物理定位仍知之甚少.该文以微卫星(SSR03)序列为探针,对家蚕染色体进行荧光原位杂交,在减数分裂的间期、粗线期、中期染色体上都观察到单一荧光信号,表明该序列为单一序列微卫星遗传标记.利用ImageJ软件测算杂交信号在该染色体上位置,信号位点距离近端的相对长度与该染色体相对长度比值为12.502±0.470.SSR03可作为家蚕染色体物理图的遗传标记.One of the Simple Sequence Repeats (SSR) ISSR03 was used as a probe for fluorescence in sltu hybridization (FISH) on meiotic chromosome preparations from B. mori females. A single hybridization signal was detected on the interphase, pachytene and metaphase chromosomes. It was thus concluded that this SSR was a single copy marker on the chromosome. The cells whose pachytene chromosomes had positive hybridization signals and whose pachytene chromosome dispersed well were selected for measurement by the software Image J. The position of the probe was located on the site at which the ratio the relative length of the distance of the signal to the acrocentric point to that of the whole chromosome was 12. 502±0. 470.
