家蚕核型多角体病毒gp64基因的定位及克隆

摘　　要：以苜蓿丫纹夜蛾核型多角体病毒ＡｃＭＮＰＶ的膜表面糖蛋白基因ｇｐ６４作为探针，对经不同内切酶酶切的家蚕核型多角体病毒基因组ＤＮＡ进行Ｓｏｕｔｈｅｒｎ杂交分析，发现ＢａｍＨⅠ酶切产生的４．２ｋｂ和７．６ｋｂ片段呈阳性杂交，经ＤＮＡ片段回收，将４．After the digestion of Bombyx mori NPV( Bm NPV) genome DNA with different restriction enzymes,the gp64 gene was localized with the gp67 gene probe from Autographa californica NPV. Two positive bands were found in the Bm NPV genome digested with Bam HⅠ, and their sizes were 4 2 kb and 7 6 kb respectively. The 4 2 kb fragment was cloned into the pBluescript vector , and the recombinant plasmid was named as pZDBM42.
