摘 要:目的研究格尔德霉素(geldanamycin,GDM)对肝细胞生长因子(hepatocyte growth factor,HGF)诱导的人胶质瘤细胞基质金属蛋白酶(matrix metalloproteinases,MMP)-2及MMP-9活性的影响。方法应用细胞培养技术培养人恶性胶质瘤细胞系U251-MG和U87-MG,用酶谱法测定MMP-2及MMP-9活性。结果HGF作用24h后,U251-MG细胞中MMP-2及MMP-9活性较正常组明显增强(P<0.05);而GDM组则能够显著抑制其活性(P<0.05),并且1000nmol/L的GDM对HGF诱导的肿瘤细胞MMP-2及MMP-9活性增强有明显抑制作用(P<0.05)。结论GDM能够抑制胶质瘤细胞MMP-2及MMP-9的表达,而且对HGF诱导的人胶质瘤细胞MMP-2及MMP-9的活性增强具有明显抑制作用。Objective To investigate the effects of geldanamyein (GDM) on the matrix metalloproteinases (MMP)-2 and MMP-9 activities in glioma cell induced by hepatocyte growth factor (HGF). Methods U251-MG and U87-MG of human malignant glioma cells were cultured by cell culture technique. MMP-2 and MMP-9 activities were examined by photodensity zymogram methods. Results MMP-2 and MMP-9 activities in U251-MG and U87-MG ceils treated with HGF for 24 h increased significantly compared with those of normal control group (P 〈 0.05), while GDM inhibited this increasing tendency obviously (P 〈 0. 05). Also 1 000 nmol/L GDM significantly inhibited the increase of MMP-2 and MMP-9 activities in these two glioma cells induced by HGF ( P 〈 0.05). Conclusions GDM could inhibit the increase of MMP-2 and MMP-9 activities in U251-MG and U87-MG cells, also significantly depress their activity increase in these two glioma cells induced by HGF.
