摘 要:目的观察p21^wafl11“‘基因转染对人乳腺癌细胞增殖活性和中心体复制的影响。方法应用脂质体介导法将pIRES—p21^wafl真核表达载体转染入人乳腺癌细胞系MCF-7,MTF法检测细胞增殖,流式细胞仪检测细胞周期分布,免疫荧光技术检测乳腺癌细胞的中心体复制情况。结果p21^wafl基因转染后乳腺癌细胞生长显著受抑制,细胞周期停滞于G1期,转染组G1期、S期和G2/M期细胞比例分别为(70.52±3.21)%、(16.38±1.17)%、(9.42±0.98)%,未转染组则为(49.84±1.58)%、(36.83±1.36)%、(15.64±1.09)%,两者比较差异有统计学意义,P值分别〈0.01、〈0.05、〈0.05。中心体复制异常的乳腺癌细胞数较转染前明显减少(P〈0.01)。结论p21^wafl基因转染能够抑制人乳腺癌细胞增殖和中心体的异常复制,进而阻止乳腺癌的发生发展,可能成为乳腺瘕治疗的一种新手段.Objective To explore the role of transfected p21^walf gene in the duplication of centrosomes and the proliferation of human breast carcinoma cells. Methods p21^walf gene packaged with lipofectin was transfected into the cells of a human breast carcinoma cell line MCF-7, which stably expressed p21^walf agene. The cell growth was detected by using MTT. The cell cycle pattern was assayed by using flow cytometry. The duplication of centrosomes of breast carcinoma cells was detected by using indirect immuno- fluorescence microscopy. Results After transfection of pIRES-p21^walf The p21^walf mRNA and protein of MCF-7-p21^walf were highly expressed as compared with MCF-7 cells. Cell growth was obviously inhibited and resulted in an accumulation of cells in G1 , presenting that the proportion of the cells in G1 phase was obviously increased from (49.84 ± 1.58 ) % to (70.52 ± 3.21 ) % , and that of the cells in S and G2/M phases was obviously decreased from ( 36.83 ± 1.36 ) % to ( 16.38 ± 1. 17 ) % and from ( 15.64 ± 1.09 ) % to ( 9.42 ± 0.98 ) %, respectively, as compared with control group. The proportion of cells which contained abnormal centrosomes in MCF-7-p21^walf was obviously decreased as compared with MCF-7 cells. Conclusion The transfection of p21^walf gene could inhibit the growth and the abnormal duplication of centrosomes of human breast carcinoma cells, which even could inhibit the genesis and development of breast cancer and might be become a new method to treat the breast cancer.
