葡萄糖调节蛋白94在人肝癌组织中的表达及其临床意义

摘　　要：目的探讨葡萄糖调节蛋白94（Grp94）mRNA及蛋白在肝癌组织和癌旁组织中的表达情况及其与肝癌发生的关系。方法肝癌患者手术切除标本10例，应用半定量逆转录-聚合酶链反应（RT-PCR）分别检测肝癌组织和癌旁组织细胞Grp94mRNA基因的相对表达量，并通过免疫组织化学SP法检测10例肝癌组织和癌旁组织标本Grp94蛋白的表达情况。结果（1）半定量RT-PCR显示：Grp94mRNA在肝癌组织中均呈高表达，相对表达量为0．7490±0．7508，而在对应的癌旁组织中呈低表达或无表达，相对表达量为0．44662±0．50000两组之间差异有统计学意义（P〈0．05）。（2）免疫组织化学及图像分析结果显示：在肝癌组织和癌旁组织中Grp94蛋白平均灰度扫描值分别为158．0800±20．5952、122．2800±24．4148两组之间差异有统计学意义（P〈0．01）。结论Grp94在肝癌中的表达明显增高，可能在肝癌的发生、发展过程中发挥重要作用。Objective To explore the expression of the glucose-regulated protein 94 （Grp94） in human hepatoeellular carcinoma （hHCC） and cirrhosis specimens and its clinical significance. Methods Ten cases of hHCC specimens were collected from the patients who received operation for liver cancer from Feb. 2005 to Jan. 2006. Semi-quantitative RT-PCR assay and immunohistoehemistry were used to detect the Grp94 mRNA and Grp94 protein expression respectively. Results Semi-quantitative RT-PCR revealed that the expression of Grp94 mRNA in the liver cancer specimens and cirrhosis specimens was 0.7490 ± 0. 7508 and 0. 446 62 ±0. 500 00 respectively （ P 〈 0.05 ）. Immtmohistoehemistry showed the level of Grp94 protein expression of the cancer specimens was （ 158. 0800 ±20. 5952 ） , significantly higher than that of the cirrhosis specimens （122.2800 ±24.4148） （P〈0.01）. Conclusion The levels of Grp94 were elevated in the hHCC specimens. Grp94 may play some roles in the pathogenesis of liver cancer.
