摘 要:研究两个反义核酸(18mer,21mer)对人白血病HL—60细胞中c-myc基因表达的抑制作用。用HL-60活细胞计数,RNA含量的RT-PCR测定和c-myc基因蛋白的免疫组化染色检查。结果:两个反义寡核苷酸都能抑制HL-60细胞的增殖,增殖抑制率为11%~75%。反义核酸还抑制c-mycmRNA和Myc蛋白的表达,而对c-mcy基因正常表达的红白血病细胞K562的增殖和PHA刺激的人淋巴细胞的增殖没有影响。结论:反义寡核苷酸能抑制c-myc基因的过度表达。In order to study the effect of two antisense oligodeoxynucleotides (18,21 mer) on the c-myc gene expression.METHODS Numbering living HL-60 cells, Measuring the level of RNA by RT-PCR and staining c-myc gene protein by immunocytochemical method. RESULTS Not only the proliferation of HL-60 cells but also the expression of c-myc gene mRNA and Myc protein were inhibited by antisense oligodeoxynucleotides. Cell inhibition rate ranged in 11%~75%. The inhibition of cell proliferation by antisense oligodeoxynucleotides was sequence-specific, dose-dependent and exposure time-related. Phosphorothioate oligodeoxynucleotides exhibited higher inhibition protency than phosphodiester oligodeoxynucleotides.CONCLUSIONS The antisense oligodeoxynucleotides can inhibite c-myc gene over expression.
