摘  要:研究维甲酸受体α(RARα)基因在乳腺癌细胞中的表达情况。用NorthernBlot、WesternBlot和免疫组化等方法。结果:RARα受体蛋白表达与其mRNA表达相一致,RARαmRNA表达在ER阳性乳腺癌细胞中比ER阴性的乳腺癌细胞高,RARα受体蛋白表达也是如此,RARαmRNA及RARα受体蛋白表达都与ER状况相关。结论:3种检测方法都具可信性,实际工作中可以因地制宜选用合适的检测手段为临床服务。Retinoic acid receptor-α (RARα) plays a major role in the growtg ubgibitory effect of retinoic acid on human breast cancer cells, maybe it could serve as an indicator to guide the treatment and prevent of breast cancer with retinoic acid in clinic. All previous researchs were based on observing the changes of RARαmRNA expression. In this study the expression of RARα was studied in mRNA level and protein level.METHODS Northern Blot. Western Blot and Immunohistochemistry were used. RESULTS RARα protein expression was correlated with RARα mRNA expression. RARα mRNA expression was higher in ER-positive human breast cancer cell lines than that in ER-negative ones. So was RARα protein expression. Both RARα mRNA and RARα protein expression were associated with ER status.CONCLUSIONS These results demonstrate that these three different methods are reliable, fitable approach can be adopted to determine the expression of RARα in human breast cancer in practice.
