大鼠脑出血后IL-1β表达及其与凝血酶的关系

摘　　要：目的观察大鼠实验性脑出血(ICH)后不同时间点血肿周围组织白细胞介素1β(IL-1β)炎性细胞表达及其与凝血酶的关系。方法免疫组化法观察ICH后不同时间点血肿周围IL-1β的表达情况及凝血酶对其表达的影响。结果在造模后1h,ICH组和凝血酶组血肿周围即可见IL-1β炎性细胞表达,6h达高峰,以后逐渐下降,48h无阳性细胞表达。在相应时间点,水蛭素组血肿周围IL-1β的表达水平均明显低于脑出血组(P<0.01),且细胞着色浅。假手术组仅有少量阳性细胞表达。结论IL-1β参与ICH后脑损伤,凝血酶可能通过促进IL-1β的表达而产生ICH后脑损伤作用。Objective To investigate the expression of interleukin-1β（IL-1β） in brain tissues after experimental intracerebral hemorrhage （ICH） in rats at different times and its effect on thrombin. Methods The expression of IL-1β in cerebral tissues at different time and the effect of thrombin on it were detected by immurohistochemitry techniques. Results There were IL-1β expressions of the perihematoma tissues found at 1 h and reached to a maximal value at 6 h after ICH and thrombin. Subsequentenly it decreased gradually and disappeared at 48 h. The IL-1β expression level of hirudin group was obviously lower than that of ICH group. Conclusion IL-1β plays a role in brain injury in ICH. Thrombin may have a brain injury role after ICH through improving IL-1β expression.
